Alkali-tolerant Protein A, Recombinant, Lyophilized
Compared with a native Protein A, Alkali-tolerant Protein A has been modified to (1) enhance the specific binding capacity to the Fc region of immunoglobulin molecules, and (2) to improve the protein stability and alkali resistance.
For efficient covalently conjugation to resins, one cysteine residue has been inserted at the C-terminus of the Alkali-torelant Protien A.
Protein A is a cell wall component produced by a number of strains of Staphylococcus aureus. It consists of a single polypeptide chain with few or no carbohydrate. Recombit Protein A is produced in E. coli and in the same manner as native Protein A. The Protein A molecule contains four high-affinity binding sites that can interact with the Fc region from IgG of a number of species. Optimal binding occurs at pH 8.2, but binding is also effective at physiological conditions (pH 7.0 to 7.6).
Protein A can be coupled to a wide variety of reporter molecules including fluorescent dyes, enzyme markers, biotin, colloidal gold and radioactive iodine without affecting the antibody binding site. Protein A can also be immobilized on a solid support, such as agarose or acrylic beads, to purify immunoglobulins / antibodies, or used in immunoprecipitation assays.
